MI-219, an HDM2 inhibitor, decreases the engraftment, self-renewal, and homing of LSCs via direct p53 stabilization and reactivation [73], and RITA, a p53-HDM2 inhibitor, in combination with CPI-203, a Bromodomain and Extra-Terminal (BET) inhibitor, decreases LSC engraftment by preventing p53 degradation in a mouse model of CML [74]. The gene discussed is TP53; the disease is chronic myelogenous leukemia, BCR-ABL1 positive.
