After validating the gastric stability and intestinal enzyme-triggeredpayload release of the developed casNP/insulin/C10in vitro using simulated gastric and intestinal fluids, in vivo studies on an STZ-induced diabetic mouse model wereperformed, which revealed a fast gastric emptying of casNP/insulin/C10 with a half-emptying time of <15 min, a quick and long-lastinginsulinization in the liver and plasma from 1 to 8 h after oral gavage,and an insulin bioavailability of 8.3%. This evidence concerns the gene INS and diabetes mellitus.
