MAPT and amyloidosis: Overall, our findings support an interpretation of the trophic level rebalance in which: (1) early amyloid deposition in DMN hubs and subsequent tau-driven neurodegeneration lead to reduce causal influence of the DMN, SAN, and SMN; and (2) less protein-burdened regions such as visual and thalamic systems become more influential, likely using the spare capacity in these networks to maintain cortico-subcortical communication.
