To check whether IGF2BP3 regulates the RAN translation of FMRpolyG we first knocked down (KD) IGF2BP3 levels using short interfering RNA (siRNA) in human embryonic kidney cells (HEK-293T), human cervical carcinoma (HeLa), and human neuroblastoma cells (SH-SY5Y) with a transient expression of RAN-translated FMRpolyG containing tract of either 16 or 99 glycine residues tagged with green fluorescent protein, GFP (FMR16xG and FMR99xG, respectively). The gene discussed is RAN; the disease is cervical carcinoma.
