This expression pattern was further confirmed by real-time PCR and immunofluorescence, which showed increased level of Ptbp1 mRNA and PTBP1 protein in the peri-infarct area after PT (Figure 1F-G) and transient middle cerebral artery occlusion (tMCAO) (Figure S1A-B). Furthermore, our examination of the peri-infarct brain tissue at day 3 post-stroke revealed a significant decrease in circSCMH1 levels, concurrent with a progressive increase in PTBP1 levels as the stroke evolved (Figure 1H).
