A recent study demonstrated multi-component synergistic killing of DLBCL cell lines by various combinations of ibrutinb (Btk inhibitor), Venetoclax (Bcl2 inhibitor), lenalidomide (promotes degradation of the IKZF1 and IKZF3 transcription factors that act downstream of the NF-κB/IRF4 pathway to promote the survival of malignant B cells), and prednisone (an R-CHOP component that inhibits the NF-κB pathway) (95). The gene discussed is NFKB1; the disease is diffuse large B-cell lymphoma.
