The results revealed that Tregs from normal individuals were predominantly located in clusters 0, 1, and 6 (overall on the left), while Tregs from ITP patients clustered mainly in groups 2–5 and 7–9 (overall on the right), as shown in Fig. 1B. Based on literature and gene expression patterns [3, 10–17], we annotated the cells into six Treg subpopulations: Annexin A1(ANXA1)high, C-C Motif Chemokine Receptor (CCR) 6high, Ikaros Family Zinc Finger 2 (IKZF2)high, Granzyme A (GZMA)high, CCR7high, and FOXP3high (Fig. 1C). The gene discussed is ANXA1; the disease is autoimmune thrombocytopenic purpura.
