Among agents that downregulate SIRT1 protein expression or activity, EX‐527, a potent and selective SIRT1 inhibitor, delivered via intraperitoneal injection, has been shown to mitigate perturbations in the activity of necroptosis‐associated metabolic enzymes downstream of ischemic injury, reduce cerebral infarct volume, and improve survival rates in models of cerebral ischemia [79].
