Multiple CNS-relevant modalities currently under development for WM deserve evaluation in dedicated BNS cohorts, including BTK degraders [47], venetoclax with documented CSF penetration [48,49], CAR-T approaches active in primary and secondary CNS lymphoma [50,51], and IRAK-4 inhibition targeting MYD88/IRAK signaling with CNS penetration [52,53]. Here, MYD88 is linked to primary central nervous system lymphoma.
