For GLP1R agonism, genetically proxied lower BMI by GLP1R variants was consistently associated with a reduced risk of obesity, with relationships observed for GLP1R agonism in the MVP BMI replication data and a directionally consistent, though less precise, estimate in the primary BMI GWAS data (Figure S4a-b, Table S3). Here, GLP1R is linked to obesity due to melanocortin 4 receptor deficiency.
