The prominent Alzheimer’s disease (AD)-associated single nucleotide polymorphisms (SNPs) are located in the BIN1 5′ region, 25 to 30 kb upstream of the coding sequence (2, 11), indicating that they reside in regulatory or enhancer regions that can confer AD risk by modifying BIN1 expression in brain cells (12, 13). Here, BIN1 is linked to early-onset autosomal dominant Alzheimer disease.
