Targeting these immunosuppressive pathways is a key focus of clinical research—PD-1 blockade is FDA-approved for recurrent metastatic HNSCC and resectable locally advanced HNSCC expressing PD1 ligand (PDL1), and inhibitors of adenosine and TIGIT blockade have demonstrated antitumor activity in preclinical studies, particularly when combined with PD1 inhibitors (3–6). Here, TIGIT is linked to head and neck squamous cell carcinoma.
