In this way, antibodies directed against conserved alpha-helices in the MUC1-C/ED have been generated for the development of (i) an allogeneic CAR T cell therapy, designated P-MUC1C-ALLO1, that has been well tolerated in the clinic, and (ii) an anti-MUC1-C (M1C) antibody-drug conjugate (ADC), which based on promising preclinical anti-cancer activity in the absence of toxicity is being advanced by the NCI NExT Program for IND-enabling studies and clinical evaluation. Here, MUC1 is linked to cancer.
