To test whether the binding of 14-3-3 proteins can suppress neuronal Tau aggregation, we expressed pro-aggregant FTD-mutant human Tau (eGFP-TauP301L/S320F) in primary hippocampal mouse neurons, which leads to spontaneous eGFP-TauP301L/S320F aggregation into NFT-like, fibrillar aggregates in a subset of neurons (~2% NFTs in DMSO control; Fig. 1a, b).
