These findings implied that the combination of metformin with Pyruvate dehydrogenase kinase 1 (PDK1) and Hexokinase-2 (HK2) (related to the glycolysis process) or NAMPT inhibitors could potentially intensify the cytotoxic effects of metformin by obstructing the metabolic rewiring capacity and potentiating the mechanism of action of metformin (associated with ferroptosis induction) of AML cells.
