PTEN and Anxiety: The homozygous germline knockout of PTEN is embryonically lethal, while heterozygous mice show behaviors resembling ASD, including deficits in social interaction, anxiety and repetitive behaviors, as well as macroencephaly, hypertrophy of individual neurons, and increased dendritic spine counts (Di Cristofano et al., 1998; Clipperton-Allen and Page, 2014; Clipperton-Allen and Page, 2015; Tilot et al., 2014; Huang et al., 2016).
