C9orf72 and amyotrophic lateral sclerosis: Although this specific effect of TMZ on Ca2+ needs to be confirmed in animal models of aging and ALS (such as stable transgenic zebrafish lines recapitulating key features of human C9orf72 and SOD1G93A ALS), the fact that, based on its preclinical efficacy, TMZ has grabbed the attention of the scientific community for the treatment of ALS (clinical trial NCT04788745) deserves a thorough examination of the possible implications of TMZ effect on Ca2+ transients, in order to better evaluate its reappraisal as a specific treatment to slow down the progression of motor symptoms in this disease.
