Following a single dose of aCD40, we observed a significant reduction in intratumoral CD103 + migratory DCs by days 5–7 post-treatment, concurrent with increased accumulation of mature, activated cDC1 and cDC2 in tumor-draining lymph nodes (TDLNs), accompanied by robust proliferation and activation of CD4 + and CD8 + T cells.
