Finally, we assessed whether the cellular toxicity of Cas9D10A-induced DNA damage could be attenuated in the same manner as the MYCN-amplified neuroblastoma cell lines by treating BT-474, NCI-H716, or NCI-H2170 cells with either rucaparib (PARPi) or calpastatin (CAST) in the presence of Cas9D10A (Supplementary Fig. 27).
