To validate this observation, we treated different human AML cell lines (n = 5) and primary patient blasts (CD34+ cells, n = 3) with a number of widely used inhibitors related to lactate/pyruvate metabolism: AZD3965 (an inhibitor of the lactate transporter monocarboxylate transporter 1; MCT1), Compound 3k (an inhibitor of PKM2, which regulates the final and rate-limiting step of glycolysis by converting phosphoenolpyruvate to pyruvate) and FX11 (an inhibitor of LDHA). This evidence concerns the gene LDHA and acute myeloid leukemia.
