In mice, TNFα signalling in the DRG was recently shown to be sexually dimorphic in an experimental autoimmune encephalomyelitis model.38 In humans, this pathway is frequently studied in circulating immune cells, and TNFα signalling here can be regulated by testosterone levels.36 Using FACS/flow cytometry on freshly isolated hDRG, we first examine the functional relevance of this pathway in the hDRG specifically: When stimulated, CD11b+ cells from hDRG release TNFα, and TNFα stimulation can alter phosphorylation levels.
