The high risk of cancer associated with sulfonylureas in our analysis aligns with the MA of cohort studies conducted by Thakkar et al. (RR: 1.55; 95% CI: 1.48–1.63).132 The observed cancer risk may be attributed to the effect of sulfonylureas on insulin levels, as these drugs stimulate the pancreas to release insulin.133 Insulin can increase cancer risk through several mechanisms.
