Additionally, we analyzed the data of GATA4 variants associated with isolated cardiac diseases from ClinVar (https://www.ncbi.nlm.nih.gov/clinvar/) and determined that only 9.09% of pathogenic/likely pathogenic variants were found to be located in the N-ZF domain; moreover, most of the variants were located in transactivation domain 1 (TAD1, 31.82%), followed by nuclear localization signal (NLS, 22.73%), transactivation domain 2 (TAD2, 13.64%), no conserved domain (13.64%), and N-ZF (9.09%) (Figure 4B).
