PLCγ isoforms bind and, upon activation by receptor tyrosine kinases or membrane-proximal cytosolic kinases of the SYK, but also SRC (proto-oncogene tyrosine protein kinase) and TEC (tyrosine kinases expressed in hepatocellular carcinoma) families (Hajicek et al, 2019), hydrolyse phosphatidylinositol 4,5-bisphosphate (PIP2) in the inner membrane leaflet, preferentially within cholesterol-rich lipid rafts (Myeong et al, 2021). The gene discussed is SRC; the disease is hepatocellular carcinoma.
