HRD tumors can benefit from PARP inhibitor therapies even without BRCA1/2 pathogenic variants, as demonstrated by the PAOLA-1 (Platine, Avastin and OLAparib in 1st Line) study, in which patients without BRCA1/2 pathogenic variants but with high GIS scores showed increased overall progression-free survival for up to 11 months.5 The gene discussed is BRCA1; the disease is hypoparathyroidism-retardation-dysmorphism syndrome.
