In trials assessing the efficacy of BRAF inhibition in combination with MEK inhibition in the treatment of melanoma, response rates of 63-69% were seen.36-39 However, in the case of CRC, the BEACON trial found that those who received a combination of encorafenib plus cetuximab (EGFR inhibitor) exhibited an objective response rate of 20%.3 In the case of biliary tract cancers, dabrafenib plus trametinib has resulted in an ORR of 51%, however, these were all partial responses in the ROAR trial.15 Prospective data for the use of BRAF inhibitors in other GI malignancies is limited. This evidence concerns the gene EGFR and melanoma.
