In trials assessing the efficacy of BRAF inhibition in combination with MEK inhibition in the treatment of melanoma, response rates of 63-69% were seen.36-39 However, in the case of CRC, the BEACON trial found that those who received a combination of encorafenib plus cetuximab (EGFR inhibitor) exhibited an objective response rate of 20%.3 In the case of biliary tract cancers, dabrafenib plus trametinib has resulted in an ORR of 51%, however, these were all partial responses in the ROAR trial.15 Prospective data for the use of BRAF inhibitors in other GI malignancies is limited.
