MEK inhibitors (trametibinib, cobimetinib, and binimetinib) have been used in combination with BRAF inhibitors to address MAPK-driven treatment resistance, with 15–20% of melanoma patients with the BRAFV600E mutation not responding, which has been attributed to the loss of the tumor suppressor genes PTEN and NF1 [42].
