In liver cancer, LACTB enhances ferroptosis by inhibiting the cystine/glutamate transporter SLC7A11/GSH/glutathione peroxidase 4 (GPX4) antioxidant pathway and activating nuclear receptor coactivator 4 (NCOA4)-mediated ferritinophagy via a p53-dependent process (Figure 3).
