We performed two positive CRISPR screens using an approximate LC50 dose of colibactin 742 (5 μM, continuous dose) or pks+ K. pneumoniae infection (multiplicity of infection [MOI] 50, 4 h infection cycles), quantifying positively enriched genes in the resultant populations (after three passages, 12–14 days) relative to a control population treated with an inactive form of colibactin (colibactin 746) or infected by ΔclbP K. pneumoniae. This evidence concerns the gene ARAF and infection.
