This has been termed ‘Targeted Augmentation of Nuclear Gene Output’ (TANGO), and has been shown preclinically to prevent SUDEP and reduce seizures in a mouse model of Dravet syndrome in which the therapy increases the expression of the sodium channel subunit, NaV1.1 [9]; a trial of such an ASO is underway in people with Dravet syndrome. This evidence concerns the gene SCN1A and encephalopathy, progressive, early-onset, with brain edema and/or leukoencephalopathy.
