CD40 agonists have also been shown to increase sensitivity to immune checkpoint blockade (18, 19), with sotigalimab and others in its class demonstrating promise when tested in combination with anti-PD1 antibodies in clinical trials for melanoma and other disease indications (14, 15, 20), including patients who have developed resistance to prior anti-PD1 therapy (17). This evidence concerns the gene CD40 and melanoma.
