Interestingly, LIPA was recently discovered to be implicated in cancer as a molecular target of a powerful therapeutic small molecule (ERX-41) for triple-negative breast cancer and potentially other solid tumors: ERX-41 induces endoplasmic reticulum stress (ERS) resulting in tumor cell death, dependently on LIPA protein-folding activities in the ER and independently of its enzymatic function, resulting in the accumulation of unfolded proteins, causing ERS [59].
