Mechanistically, besides supplying energy to cancer cells, FASN exerts its pro-tumor effects through its positive feedback loop with HER2 (Menendez et al., 2021; Kumar-Sinha et al., 2003; Vazquez-Martin et al., 2008) as well as its interaction with nuclear factor-κB (NF-κB) (Liu et al., 2013; Wu et al., 2016), which affect downstream signaling pathways and cellular processes critical for cancer cell survival and resistance to treatment.
