Perforin released by CAR‐T cells induces pore formation in tumor cells, enabling granzyme A and B to enter, hydrolyzing gasdermin B or activating caspase 3 to cleave gasdermin E. The N‐terminal gasdermin fragment forms a pore in the plasma membrane, leading to leakage of damage‐associated molecular pattern (DAMPs), like ATP and heat shock proteins, and cytokines.
