Day seven generated CD14+ TAMs expressed both CD163 and CD206, similar to freshly isolated patient-derived TAMs, whereas PBMC-derived monocytes without tumour co-culture did not express either CD163 or CD206; moreover, during TAM generation, CD14+ cells became more granular, consistent with a pro-tumour phenotype, as assessed by an increase in side-scatter (SSC) upon flow cytometry acquisition (Figure 2A).
