Filgotinib—the newest member of the JAKi class—exerts its therapeutic effects by selectively targeting and inhibiting the kinase activity of JAK1 [7], disrupting the intracellular signaling cascade initiated by key cytokines (e.g., interleukin-6 (IL-6) and type-I interferons (IFN1), which are integral to the inflammatory processes involved in the pathophysiology of RA (Figure 1).
