Previous findings have indicated that TRPV4 channel activity increases endothelial Ca2+ levels to drive vasorelaxation.18,42,43 Given our observations that high levels of TRPV4-mediated activation reverse vasorelaxation, we speculated that endothelial Ca2+ levels might decrease at higher GSK concentrations in hypertension.
