Functional experiments were performed to modulate the methDNA status in four melanoma cell lines and demonstrate the correlation between the expression and methDNA of SLC22A17. In particular, the SK-MEL-23, WM115, A375, and SK-MEL-28 cell lines were treated with the demethylating agent 5-Aza at concentrations of 1.9 μM for SK-MEL-23, 0.75 μM for WM115 and A375, and 3.5 μM for SK-MEL-28, while the M14, A2058, and MeWo cells were excluded from 5-Aza treatment due to the low methDNA status of the upstream/downstream promoter and body hotspots (Fig. 7 and Supplementary Fig. S1). The gene discussed is SLC22A17; the disease is melanoma.
