Endothelin receptor antagonists (ERAs) represent another pharmacological treatment proven to reduce the rate of kidney function decline in type 2 diabetes and IgA nephropathy.3,4 The phase 2B Zibotentan and Dapagliflozin for the Treatment of CKD (ZENITH-CKD) dose-finding trial showed that the addition of the highly selective ERA zibotentan in combination with the SGLT2 inhibitor dapagliflozin also significantly reduced albuminuria compared with dapagliflozin alone, suggesting complementary mechanisms.5 The gene discussed is SLC5A2; the disease is chronic kidney disease.
