To demonstrate a dependency upon Notch1 for enhancer looping and the direct interaction between the MYC and NDME loci, we first performed 3C-PCR to determine whether the formation of N1ICD transcriptional condensates bring these two distal genomic loci into close physical proximity with one another in both T-ALL cells, in which this interaction has been previously described, and in HEK293 cells, to determine whether MYC super-enhancer looping occurs in this cell type as well (Fig. 7C)68–70. The gene discussed is MYC; the disease is acute lymphoblastic leukemia.
