MAPT and Alzheimer disease: Although this was not directly tested in this work, among several potential pathways, cSVD has been associated with inflammation in and around the perforating arterioles and capillaries and has been shown previously to relate to neuroinflammatory processes and astrogliosis.38,39 Such processes, in turn, have been proposed to mediate the relationship between amyloid and tau.40 cSVD-related neuroinflammatory processes may, therefore, constitute a co-pathology that triggers an increase in inflammatory markers and thus promote the amyloid-dependent pathologic processes, typical of AD.
