According to the somatic mutations reported in the 15 public datasets included, the most commonly mutated genes in CRC are TP53, APC, KRAS, PIK3CA, FBXW7, SMAD4, and BRAF. Log-rank analysis revealed that among these seven genes, KRAS and FBXW7 were not prognostically related, whereas the mutational status of the remaining five genes, TP53, APC, PIK3CA, SMAD4, and BRAF, correlated significantly with prognosis. Here, SMAD4 is linked to colorectal carcinoma.
