To investigate whether the DS variant-promoted activation of NFκB could affect the phosphorylation of MLKL that was induced by these glycans in breast cancer cells, we used immunofluorescence to determine the level of phospho-MLKL in cultures that had first been preincubated for 3 h with cardamonin, which is an inhibitor of this transcription factor [17,18,19], and then exposed to cardamonin in combination with an individual DS variant for 3.5 h (Supplementary Figure S2).
