SF3B1 and myelodysplastic syndrome: The second-generation XPO1 inhibitor, eltanexor, appears to have increased efficacy and tolerability in a small phase 2 study of MDS [32, 34]; however, despite the promising efficacy of XPO1 inhibition in SF3B1-mutated MDS, maximizing efficacy and minimizing toxicity remains a challenge, which forms the rationale for investigating combination therapies.
