PIKFYVE and cancer: Small-molecule inhibitors of the PIKFYVE have been shown to disrupt lysosome homeostasis, which involves all three types of autophagy in mammals [macro-autophagy, micro-autophagy, and chaperone-mediated autophagy [24], henceforth simply termed autophagy], as well as endo-lysosomal trafficking pathways, suppressing many critical nutrient recovery and energy production pathways in PIKFYVE-dependent cancer cells.
