Mutations in TREM2 and DAP12 geneshave been linked to an increased risk of developing neurodegenerativediseases such as Alzheimer’s disease.16 Understanding the molecular mechanisms underlying the TREM2 signalingpathway and associated signaling adaptor molecules such as DAP12 iscritical for the development of new therapeutic strategies for neurodegenerativediseases.17,18 Further research on this pathway will providevaluable insights into the regulation of microglial function and thepotential development of new drugs targeting neurodegenerative diseases. The gene discussed is TYROBP; the disease is early-onset autosomal dominant Alzheimer disease.
