In vitro cell culture studies show that all activating BRAF mutants are RAS-independent (Yao et al., 2015): neither RAS inhibition (Yao et al., 2015) nor EGFR inhibition (Corcoran et al., 2012; Prahallad et al., 2012) was able to inhibit mutant-BRAF-induced ERK phosphorylation in BRAF-mutant human CRC cell lines.
