Gain-of-function mutants of NLRP3, which are the cause of the auto-inflammatory disorder cryopyrin-associated periodic syndrome (CAPS) [87], cannot interact with CARD8, suggesting that the lack of NLRP3 inhibition by CARD8 is the reason for the inflammatory phenotype in CAPS patients [86].
