At the functional level, in vitro differentiation of GIC19 shM7B led to a decreased number of cells expressing the astrocytic marker GFAP, as well as an increased number of cells expressing the neuronal differentiation marker TUJ1, at the end of the differentiation process (Figure 6C), and histological analysis of GICshM7B-derived xenografts confirmed the lower expression of the astrocytic markers SOX2 and GFAP as well as diffuse expression of the neuronal marker SYNAPTOPHYSIN in the METTL7B-deficient tumor cells (Figure 6D).
