A durable clinical benefit of monotherapy with daily sotorasib (administered orally at a dose of 960 mg) was confirmed in the phase 2 CodeBreaK 100 trial, showing a 37.1% ORR, median PFS of 6.8 months, and median overall survival (OS) of 12.5 months in KRAS p.G12C-mutant advanced NSCLC patients previously treated with standard therapies (Table 1) (32).
